Both LaLiga and Serie A will play officials matches in Australia and the US respectively before the end of the year.
Khan, who also owns the NFL's Jacksonville Jaguars, told talkSPORT: "Why are we doing what we’re (NFL) doing? Because it’s a very American-centric sport.
"It’s a huge cultural phenomenon in the US, it’s saturated. For us to have the growth, we needed international.
"But when you look at the Premier League, it’s already an international sport. As I said earlier, by far the most competitive league in the world.
"So I would then question, what is the need for it? Where the game is played, it has (a) huge amount of international following and it’s competitive. So I would question really the need.
"Looking at it, what the NFL needs and what the Premier League needs, they’re two different things."
