In a blistering start, Adelaide almost had an early penalty when Alemao skipped past Glory skipper Jacob Burns on the edge of the area.
Burns could only put a foot out in hope but the home side's protests were waved away.
The game was being played at a frenetic pace early on with neither goalkeeper really being tested.
On the half-hour mark, Perth Glory's Chris Coyne had a hold of Robert Cornthwaite's shirt in the area after a free-kick was sent in and the referee pointed to the spot for a controversial spot kick.
Captain Travis Dodd made no mistake, sending Tando Velaphi to his right and hitting the back of the net to give the Reds the lead.
The scoreline remained that way until half-time with both sides having to make changes due to injuries - Lucas Pantelis for Adelaide and Jamie Coyne for Perth.
Moments after the re-start, Adelaide's Adam Hughes found himself at the back post with a free header from the six-yard box but completely miscued, letting the visitors off enormously.
For the remainder of the contest, Glory had enough play to get themselves back into it but the Reds were able to hold sway and grab themselves a very handy three points to kick off season 2009/10.
